In this poster, we demonstrate how Wieslab® pathway-specific functional Complement ELISAs can easily be used to measure the potency of complement inhibition of any of the complement factors involved in the cascade.
For every biological active drug there is a need for a relevant functional assay, a bioassay. The ideal bioassay is based on the mechanism of action (MOA), and can be utilized for several purposes, like efficacy and potency measurements, and release testing in CMC.
In this poster, we demonstrate how Wieslab® pathway-specific functional Complement ELISAs easily can be used to measure the potency of complement inhibition of any of the complement factors involved in the cascade.
The pathway-specific ELISAs are especially useful for assessing potency of drugs targeting the early complement factor, while inhibition of the terminal pathway can be measured with any of the complement ELISAs.
Furthermore, as none of the functional ELISAs can detect inhibition of the C5a/C5aR signal induced by the highly potent anaphylatoxin C5a formed during complement activation, we enable potency measurement of drugs targeting C5, C5a, or the C5aR1 by using our in-house developed assay ready C5a responsive cell-based reporter gene cell line.
