iLite® AAV Responsive Reporter Assay Ready Cells

BM6100

A luciferase-based reporter cell line designed to quantify AAV vector transduction efficiency and detect neutralizing antibodies that impair gene delivery—ideal for immunogenicity testing for systemic and muscle-directed gene delivery.

  • Firefly luciferase expression driven by AAV transduction
  • High sensitivity and large dynamic range
  • "Assay-ready" format for rapid and convenient use
  • Reproducible and biologically relevant results
  • Reduced assay variability and higher reproducibility compared to cells in culture
  • Normalization of cell counts and serum matrix effects using a second reporter gene

Product Overview

iLite® AAV Responsive Reporter Assay Ready Cells are based on the human embryonic kidney 293 (HEK293) cell line and are ideal for neutralizing antibody detection studies. Designed to respond to the transduction (interaction, uptake, and nuclear effects) of AAV vectors made with iLite® AAV Packaging Assay Ready Cells.

How it works

arrow
AAV packaging cell

A two-cell line setup

A packaging cell generates AAVs. We offer many different kinds of packaging cells, each producing a different AAV serotype.

AAV iLite cell

A two-cell line setup

An AAV-responsive reporter cell expresses Firefly luciferase in response to AAV transduction that can be measured using a standard luminometer. In the presence of neutralizing antibodies, the AAV transduction is impaired, resulting in a lower signal. A constitutively expressed second Renilla luciferase reporter gene can be used as an internal control of matrix effects.
