Complement C4d Flexi

The purpose of the Complement C4d Flexi reagents is to specifically detect the short linear C4d neo-epitope (both A and B genetic variants) exposed after activation of C4 and subsequent cleavage of C4b. The Complement C4d Flexi application note and reagents enables users to independently set up, customize and optimize a Complement C4d Flexi assay based on Svar Life Science’s Complement C4d assay on Gyrolab® (Gyros Protein Technologies) automated platforms or other platforms of their choice. The Complement C4d Flexi reagents may also be used for other applications of your choice.

It is recommended to set up Complement C4d Flexi assays as 3-step sandwich assays with the capture and detection antibodies used in their designated functions to ensure optimal specificity for the C4d neo-epitope in EDTA plasma samples.

APPLICATIONS for the Complement C4d Flexi Portfolio

The application note for the Gyrolab® system is developed for measuring C4d in human plasma. The C4d protein is a degradation product of the classical and lectin pathways and is increasingly recognized as an immunologic footprint of complement activation and antibody activity. It serves as a potential biomarker for diseases where antibodies can cause tissue damage, such as systemic autoimmune diseases.

C4d has the potential to detect patients at risk for the consequences of antibody-mediated disease and is also used during the development of new therapeutics that block complement activation to identify and monitor patients who may possibly benefit from these drugs.

Together with Gyros Protein Technologies, Svar Life Science has developed an automated immunoassay for the specific detection of Complement C4d in EDTA plasma samples using commercially available reagents (Complement C4d Flexi).

A new application for the Complement C4d assay to be used on the Gyrolab® automated platform from Gyros Protein Technologies has been established with an assay setup comparable to Svar’s Complement C4d ELISA (COMPL C4d RUO), using a highly specific C4d neo-epitope monoclonal antibody as capture antibody. The assay is a high performance nanoliter-scale sandwich immunoassay approach offering the following advantages:

  • Automated solution
  • Customizable
  • Rapid time to result: Automates assay workflow with results in 1 hour

Using an affinity flow-through format, Gyrolab technology simplifies assay workflows by eliminating incubations and shortening run times.

 

The proprietary CD technology, engineered with highly reproducible nanoliter microfluidics, integrates with Gyrolab platforms, which automate immunoassays with parallel processing using laser-induced fluorescence detection. This is possible through precise, automated control of centrifugal and capillary forces which steer liquid flow through nanoliter-scale microfluidic structures contained within the CD in which the assay workflow is automated.
