The Svar Life Science Complement Flexi Solutions allow users to independently set up, customize and optimize assays based on existing Svar Life Science assays. In collaboration with Gyros Protein Technologies, Svar has developed application notes for use of the Complement Flexi Solutions on the automated Gyrolab® platforms. The Flexi Solutions can of course also be used in manual assays as well as for other laboratory techniques, such as immunoblotting and immunohistochemistry, where your imagination is the only limit.
The Complement Flexi product portfolio consists of the assay components required to setup the Svar Life Science assays as 3-step sandwich assays, which is the recommended assay format and is comparable to the ready-to-use assays also provided by Svar.
The purpose of the Complement TCC Flexi reagents is to specifically detect the soluble terminal complement complex (TCC, also known as sTCC or sC5b-9) which is an end product of complement activation of all three complement pathways. The Complement TCC Flexi application note and reagents enables users to independently set up, customize and optimize a Complement TCC Flexi assay based on Svar Life Science’s Complement TCC assay on Gyrolab® (Gyros Protein Technologies) automated platforms or other platforms of their choice. The Complement TCC Flexi reagents may also be used for other applications of your choice.
It is recommended to set up Complement TCC Flexi assays as 3-step sandwich assays with the capture and detection antibodies used in their designated functions to ensure optimal assay performance.
The Complement TCC Flexi application note for the Gyrolab® system is developed for measuring TCC in human plasma. TCC is the end product of the terminal pathway and can be a result from all three complement activation pathways. Since TCC reflects activation to the end of the final terminal pathway irrespective of the initial pathway involved, it is a particularly good candidate for general evaluation of complement activation.
Increased levels of TCC can be detected in for example hemolytic uremic syndrome (HUS), Systemic lupus erythematosus (SLE) and rheumatoid arthritis (RA). TCC levels can also be used to assess complement activation in response to artificial surfaces and biomaterials in medical devices.

The Complement C4d Flexi application note and reagents enables users to independently set up, customize and optimize a Complement C4d Flexi assay based on Svar Life Science’s Complement C4d assay on Gyrolab® (Gyros Protein Technologies) automated platforms or other platforms of their choice. The Complement C4d Flexi reagents may also be used for other applications of your choice.
It is recommended to set up Complement C4d Flexi assays as 3-step sandwich assays with the capture and detection antibodies used in their designated functions to ensure optimal specificity for the C4d neo-epitope in EDTA plasma samples.
The application note for the Gyrolab® system is developed for measuring C4d in human plasma. The C4d protein is a degradation product of the classical and lectin pathways and is increasingly recognized as an immunologic footprint of complement activation and antibody activity. It serves as a potential biomarker for diseases where antibodies can cause tissue damage, such as systemic autoimmune diseases.
C4d has the potential to detect patients at risk for the consequences of antibody-mediated disease and is also used during the development of new therapeutics that block complement activation to identify and monitor patients who may possibly benefit from these drugs.
